Après s'être séparés pendant quelques années, Scooby-Doo, Fred (Freddie Prinze Jr.), Daphne (Sarah Michelle Gellar), Shaggy (Matthew Lillard) et Velma (Linda Cardellini) sont convoqués individuellement sur Spooky Island pour enquêter sur une série d'incidents paranormaux survenus dans ce lieu ultra-branché où passent leurs vacances de printemps des étudiants envahis par d'étranges monstres démoniaques.

Having been broken up for a couple of years, Scooby-Doo, Fred (Freddie Prinze Jr.), Daphne (Sarah Michelle Gellar), Shaggy (Matthew Lillard) and Velma (Linda Cardellini) are individually summoned to Spooky Island to investigate a series of paranormal incidents at the ultra-hip Spring Break hot spot; occupied by college students and the eeriw demonic monsters possessing them.
