Title: Tools and Approaches for Nontargeted Metabolomics Studies of Disease in Human Cohorts
Abstract: The potential of nontargeted metabolomics to detect and characterize early metabolic changes in subclinical disease has important implications for discovering pathophysiologic mechanisms and disease markers. Our lab has developed software tools and comprehensive LC-MS metabolomics methods that are suitably robust and informative to conduct large studies of biological specimens from human cohorts. Our work has included discovery of plasma metabolic signatures that indicate future risk of developing diabetes, cardiovascular disease, gastrointestinal cancer, and other conditions. Using examples from recent projects, this presentation will highlight approaches to acquiring nontargeted LC-MS metabolomics data, data processing and management using in-house developed tools, and approach and challenges associated with identification of unknowns.
