CUT&RUN (Cleavage Under Targets & Release Using Nuclease)
This epigenetic method is used to investigate the genome-wide distribution of various chromatin-associated proteins and their modifications. CUT&RUN is a derivative of chromatin immunocleavage (ChIC). CUT&RUN is similar to chromatin immunoprecipitation (ChIP), in that it utilizes an antibody to target chromatin-associated marks and proteins, but requires less sample material and less sequencing depths than ChIP.
Unlike ChIP, CUT&RUN does not require crosslinking of the protein to the DNA, eliminating the introduction of artifacts. CUT&RUN is a valuable tool for studying chromatin-associated proteins such as transcription factors or histone modifications genome-wide.
Ideal for: Post-docs, RAs or grad students new to epigenetics research or studying the role of transcription factors and other DNA binding proteins on gene regulation
Workshop Objectives:
- Hands on lab training with Active Motif’s CUT&RUN Kit
- In-depth protocol review highlighting challenging steps and how to overcome them, QC steps required, and trouble shooting scenarios.
- Data analysis training using CUT&RUN raw example data set / FASTQ files
Agenda:
- January 28th 1:30pm - 4:30pm Introduction lecture, wet lab using real samples
- January 29th 9:00am - 1:00pm: Wet lab continued from day 1(through library prep) using real samples, raw data analysis bioinformatics presentation and Q&A with example data set.
Required: Must bring your own pipets and tips and plan to start at 9am
